Affinity purified chimeric human-mouse monoclonal anti- collagen type II (COL2A1) antibody clone CB20 in PBS, produced in HEK293 cells. The CB20 antibody1,2 recognizes the C1 epitope on triple helical collagen II. The CB20 antibody tolerates the citrullination of Arg360, but citrullination of Arg365 abolishes binding3


Description

The CB20 antibody may be used for in vivo binding studies to locate the C1-epitope in tissues or for immunohistochemistry (IHC) with ex vivo cryofixed tissues, e.g. on collagen in the joints4. The CB20 antibody has been used to specifically block C1-epitope specific antibodies in serum5.  


The CB20 antibody, provided as human IgG1, is ideal for staining tissue sections from other species such as experimental mice and rats, where anti-human IgG specific reagents may be used for detection without cross reactivity to the IgGs in the sample.

Epitope

Mouse collagen II triple helical C1 epitope (position 359-369), with sequence ARGLTGRPGDA6, with binding focus on the latter Arginine7.

Specificity

The CB20 antibody recognizes the conformational C1 epitope on the triple-helical structure of the native CII molecule.

Storage

Centrifuge briefly prior to opening vial. Store at +4 °C short term (1-2 weeks). Aliquot and store at ‑20 °C long term. Avoid repeated freeze/thaw cycles.

Recommended dilution

It is recommended that the user determines the optimal dilution for their application. The typical starting working dilutions are: Functional assays (cell culture) 1:200-1:500, IHC 1:1000.
